Bit of a dump on what each character is going through:

Lupin: Vampire - bitten by a vampire but managing to regain control, Lupin's goal is to defeat the vampires that bit him in the first place, but that doesn't mean he can't use his new perks to steal some things.

Jigen: Werewolf - after an incident when stealing an artefact, The Fang of Gévaudan, Jigen transforms whenever the clock strikes midnight.

Goemon: Vessel, Medium - Having many cursed individuals and items around him, Goemon's gains the ability to see into the next world. With visions of Ghosts and sometimes being possessed by his ancestors, Goemon hopes this otherworldly knowledge can aid them in returning to normal.

Fujiko: Siren - a mermaid amulet gives Fujiko the ability to swim underwater, unfortunately she gets the urge to sing and lure sailors to their death. Lupin and Goemon often find themselves under her song and poor Jigen has to put a stop to it.

Zenigata: 'Hunter' - after noticing the gang having new and unbelievable abilities he changes his tactics and makes himself a Van Helsing styled hunter. It plays out like you'd imagine, with garlic, crucifixes, crossbows, silver bullets and a spray bottle of Holy Water!
